We’re thrilled to announce two features to the TokuProfile Dashboard for multiparameter cytometry datasets: “p-values on demand” and the ability to assess the frequency of any cell population as a percent of any preceding ancestor

P-values on demand

With this Tokugawa 1.0 release, the Toku Dashboard instantly returns a p-value for any valid comparison—including between responders and non-responders, time points, or doses. For multiple comparisons (e.g., frequencies of unique immune cell populations between responders and non-responders), p-values can be ranked and arranged in ascending order to highlight which comparisons are statistically significant.

This release also becomes useful when you need to include or exclude specific samples from a comparison. For instance, if you notice an outlier that should be removed, hover over the data point to view the sample name. Open the Filter drop-down, click Select All, then uncheck the outlying sample. The p-value recalculates instantly based on the updated selection.

Hmm, p-values—what are those again? It’s easy to forget the stats when you’re not thinking about them every day. When that happens, just hover over the p-value text. A tooltip will pop up with a quick refresher on how the statistic was computed.

Ancestor-based calculations

When looking at immune cell frequencies, it’s important to calculate them as a percentage of the right ancestor or top-level population. For example, if you’re interested in CD4+ central memory T cells, it’s much more meaningful to view them as a percentage of total CD4+ T cells or total T cells. 

Before this release, that wasn’t possible—you could only see them as a percentage of all non-granulocytes. Now, you can display frequencies using any ancestor in the cell lineage. This makes it easy to tailor your analysis to the specific questions of your drug program.
